  Abstract:

Magnetic nanoparticles are considered to have great potential in biomedical applications due to their intrinsically peculiar magnetic, high efficiency catalytic activity and good biocompatibility. This presentation will focus on the development of magnetic nanoprobes and application in imaging-guided cancer precision therapy in our group recent years. Firstly, we try to utilize the physical properties of magnetic nanoparticles to improve the diagnosis and treatment for cancer. A novel multifunctional probe Fe5C2-ZHER2:342 was designed for magnetic resonance imaging/photoacoustic tomography-guided targeting photothermal therapy.[1] Furthermore, Fe5C2 NPs can also be a remote-controlled platform for photochemothermal cancer therapy by loading anticancer drug doxorubicin. Next we have developed a magnetic nanoprobe based on pH-sensitive Fe5C2@Fe3O4 nanoparticles for switchable magnetic resonance imaging-guided cancer therapy by reactive oxygen species. To achieve multi-modality imaging, recently, multifunctional heterostructure Au-Fe2C-ZHER2:342 were fabricated as a multifunctional nanoprobe for triple-modal magnetic resonance imaging/multispectral photoacoustic tomography/computed tomography-guided photothermal therapy. Moreover, to effectively utilize the ROS effect, we proposed magnetic nanoprobe of near-infrared light and cancer microenvironment dual responsive size-switchable DOX-ICG@Fe/FeO-PPP-FA nanocapsules with PLGA which offers fluorescence imaging/magnetic resonance imaging-guided chemotherapy, photothermal therapy and chemodynamic therapy, particularly, the degradation of PLGA matrix and Fe/FeO NPs was observed, significant phenomenon for further medical applications. In summary, it is expected that magnetic probes are promising platforms which provides imaging-guided therapy triggered from their intrinsic physical properties and the contribution of other counterparts such as targeting molecules and drugs.
